The C&L Dye Fluorometer

C&L Dye Fluorometer and Olympus IX70 Microscope

The C&L Dye Fluorometer is shown here with an Olympus IX70 inverted microscope.  It utilizes unique microprocessor-controlled filter wheels for wavelength selection in the illumination and detection optics.  The filter wheels can be programmed for excitation and emission for numerous types of  applications.  With this fluorometer, you do not have to settle on measuring one or two wavelengths. The C&L Dye Fluorometer can measure up to eight excitation-emission wavelength pairs in rapid succession.  In fact, it's the fastest multiwavelength fluorometer available, bar none.  

The Dye Fluorometer connects to your fluorescence microscope using input and out adapters.  The output adapter contains optics and a calibrated field-stop iris to limit the area of detection to any centered circular area within the observed field. Using this feature, fluorescence from the entire field or from a single cell can be collected and measured.  Without a microscope, it can also attach to a Cuvette Module.

The Dye Fluorometer is controlled by FluorMeasureTM, a powerful Windows-based software application.  The software provides all that you need for single, ratio or multiwavelength fluorescence measurements.  

1. Model PC-DAQ Controller

The C&L Dye Fluorometer is operated by the Model PC-DAQ Controller.  This is an PCI bus PC card (not shown) which is easily installed in your computer workstation.  The card is completely Plug and Play compatible and has an accompanying breakout box (in forefront of above image). The PC-DAQ Controller operates the Model S48D Illumination Source and Detection Module and controls all aspects of data acquisition using Windows-based software. The data acquisition software graphs incoming data and ratio data in real time. Single and multiwavelength protocols can be set up with a minimum of effort and all the setting can be recalled for later use.  Up to eight analog data channels can also be acquired (0 to 5 Volt), monitored in real time and saved together with your fluorescence data. 

A separate Data Analysis software package is used for analysis.  The user can examine the data in fine detail and apply several signal processing functions, including ion concentration calculations, Fourier transform/frequency domain analysis, digital filtering (high pass, low pass, notch and passband), and data smoothing (Savitsky-Golay). The data can also be exported for incorporation into other software programs.

The Model PC-DAQ Controller may also be purchased separately, for use in analog data collection or photon counting applications.

2. Model S48D Illumination Source

The Model S48D Illumination Source for fluorescence excitation consists of a xenon arc lamp and an integrated filter wheel for wavelength selection. This illumination source can hold either a 4 or 8 position filter wheel.  The 4 position wheel uses 1 inch diameter filters, while the 8 position wheel uses 1/2 inch diameter filters.  The filter wheels are easily changed, as are the individual filters in the wheels.

This illumination source uses either a 150W or 300W Xenon arc lamp.  The lamp is self-aligning, so that replacing the lamp is a simple task.  The illumination source outputs light using a quartz liquid light (5 mm diameter) and has an output range from approximately 320 to 700 nm.  The Model S48D also incorporates a variable light attenuator so that the illumination intensity can be varied precisely over a 100 step range without affecting the "color" of the output.

This illumination source may also be purchased as a separate component for use in other systems.   It can be used as an illumination source for fluorescence video imaging systems. To interface this source with hardware from other manufacturers, contact us.  The Model S48D Illumination Source, when purchased separately, can be controlled with the Model PC-DAQ Controller card.

3. Model D48 Detector Module

The Model D48 Detector Module contains an integrated filter wheel for wavelength selection. When used in combination with the Model S48D Illumination source, it provides the complete optics for fluorescence excitation and photon detection.  This detector can hold either a 4 or 8 position filter wheel.  The 4 position wheel uses 1 inch diameter filters, while the 8 position wheel uses 1/2 inch diameter filters.  The filter wheels are easily changed, as are the individual filters in the wheels. 

The detector incorporated a sensitive end-on photomultiplier tube operating in the photon count mode for unsurpassed sensitivity. Multialkali and bialkali versions are available.  The detector also includes a shutter.

The Model D48 Detector Module may also be purchased separately to serve as stand-alone multiwavelength fluorescence detection system.  It can be added to an existing imaging system to provide faster and more sensitive fluorescence detection.  In this application, data gathered using this detector with the Model PC-DAQ Controller is immediacy available without time-consuming image analysis.
